Product Description

Serum contains proteins, amino acids, glucose, hormones, etc., among which proteins are mainly albumin and globulin. There are various types of amino acids, which are the basic components of protein synthesis in cells. Some of these amino acids cannot be synthesized by animal cells themselves (known as essential amino acids). The hormones contained in serum include insulin, growth hormone, and various growth factors (such as epidermal growth factor, fibroblast growth factor, insulin-like growth factor, etc.). Serum also contains various unknown factors that promote cell growth, adhesion, and other active substances, which can promote cell growth, proliferation, and adhesion.

Each batch of raw blood material from Li Ji Biology's fetal bovine serum has undergone strict screening and has been filtered through three consecutive layers of 0.1 μ m filter membranes. The filter material meets the requirements of the US FDA regulations. The entire production process complies with international fetal bovine serum production standards and undergoes strict quality control and analysis and testing of physical, chemical, and microbiological indicators. 1. This product is only for scientific experimental research and should not be used in clinical diagnosis, treatment, or other fields.

2. Thawing and Storage of Serum: After taking out the serum from a -20 ℃ or lower temperature freezer, it is recommended to thaw it in a 4 ℃ freezer. Gently shake it evenly every once in a while, and do not directly thaw it at a higher temperature to avoid protein aggregation and precipitation caused by significant temperature changes, which can lead to a decrease in serum quality. If a bottle cannot be used up in the short term, it is recommended to pack it aseptically and then put it back in the freezer to avoid repeated freeze-thaw cycles. After long-term storage at 4 ℃, the denatured lipoprotein and fibrin in the serum form flocculent substances that deteriorate.

3. Serum inactivation: Heating can inactivate the complement system in serum. In rare cases (when culturing ES cells and smooth muscle cells), inactivation is recommended. Serum inactivation is not recommended when culturing other cells. Serum inactivation is very prone to precipitation. If inactivation is necessary, please operate according to the principle of 56 ℃, 30 min, and slight shaking. High inactivation temperature, long time, and uneven shaking are all prone to precipitation.

4. Serum free culture medium: Serum free culture medium has advantages in repeatability of results and in vitro cell differentiation, but serum is still the most basic additive in the culture medium, especially in primary culture or when cell growth is poor. Serum containing culture medium is often used for cultivation first, and then replaced with serum-free culture medium when cell growth is vigorous.

      A:胎牛血清在4℃的条件下,一般可以保存6-12个月。但这并不是绝对的,实际的保存时间会受到多种因素的影响,包括血清的质量、容器的密封性、冰箱的稳定性等。为了保证血清的质量,建议尽量在购买后3个月内使用,如果需要长期保存,最好冷冻在-20℃或更低的温度下,这样可以保存1-2年甚至更久。但请注意,每次取用时应避免反复冻融,以防止蛋白质和营养成分的损失。在使用前,应详细阅读产品说明书,并按照供应商的建议进行操作。

      A:①当储存于2至8℃的条件下,血清中可能存在的纤维蛋白、脂蛋白、冷凝集素、玻连蛋白等会发生聚集,并且形成肉眼可见的沉淀或混浊。通常并不会影响血清的使用性能。如果需要去除沉淀物,可通过离心(400g,5分钟)去除沉淀后使用上清液。②其形成常见原因如下:温度剧变(如从-20℃直接移至37℃解冻)、反复冻融、热灭活(56℃处理)及4℃或室温长时间放置等因素影响。为减少沉淀,建议在4℃缓慢解冻(期间轻轻摇晃匀)、分装保存避免反复冻融、尽量避免对血清进行热灭活(易出现沉淀物或沉淀物增加)。

      Q:血清是否需要进行热灭活操作呢?

      A:依据细胞的来源或者种类。多数昆虫细胞系与胚胎干细胞系都需要应用热灭活的血清。如果需要进行热灭活,推荐使用56°C水浴30分钟(期间轻轻摇显匀)的方法。这种方法可以在一定程度上保留血清中的营养成分,同时有效去除补体。

      Q:对于一些比较敏感细胞比如原代神经元细胞,更换血清和胰酶的注意事项?

      A:以原代神经元细胞为例,其通常较为敏感,更换培养基血清时,建议采用梯度更换法,例如起始阶段使用25%新血清与75%旧血清混合,逐步提高新血清比例。更换消化酶(如胰酶)时,应适当降低酶浓度,以减轻对细胞的刺激损伤。此外,根据具体细胞类型和研究需求,可能需要补充特定的营养物质或生长因子。建议查阅相关文献以确定最适宜的补充方案。

      Q:选择合适血清,对养原代细胞的影响?

      A:在培养原代细胞时,需注意其细胞群体中分裂能力的异质性。培养条件(越接近体内微环境)越适宜,分裂能力强的细胞(如祖细胞)越容易占据优势,从而增加可传代的次数;反之,若终末分化细胞占主导,则传代能力有限。
